The cheapest is Thai Airway, stop-over in Bangkok. Add in all the surcharges is abt $900++.

Next will be Malaysia Airlines, stop-over in KL. Add in all the surcharges is abt $1,400++.

Then come the SIA, direct flight. I have checked the SIA website, depend on how u search the website, it's range from abt $1,300++ to abt $1,500++ with all surchagres.

Not sure abt ANA and Japan Airlines though...u may want to search for yourself.

And also to really enjoy the cheapest rates, it's always good to book yr flight early. E.g., 2-3 mths in advance. And also have to depend on the dates that u are going, whether they are within the peak periods.

Generally, there is lesser competition flying to Osaka than Tokyo for the airlines. As such, it is tougher to find cheap airfare to Osaka as compared to Tokyo.

The non-direct flights are usually cheaper in this aspect - Thai Airways, Malaysia Airlines and Cathay Pacific. These are usually going for around $450 to $600 (taxes excluded). Do note that Osaka airport imposes a slightly higher airport taxes as compared to Tokyo Narita Airport. The taxes and surcharges usually can come up to around S$250 to S$300.

ANA, Japan Airlines and Singapore Airlines do have a special fare of around $600 to $650 (exclude taxes) if there are 4 passengers flying together throughout the trip. Japan Airlines and SQ flies directly to Osaka while for ANA, it requires you to change to the domestic flight from Tokyo to Osaka and vice versa. If there are only 2 passengers flying together, the fare on these 3 airlines are around $800 (excl taxes). Flying alone will usually attract a fare of around $1,100.

Alternatively, you may wish to consider taking Northwest Airlines for around $400, based on the current promotion price (excl taxes) and buy the 7-day JR pass (cost around $400, depending on exchange rate) which allows you to take the JR trains and the Shinkansen (Bullet Train) to Osaka. With the bullet train, you should be able to arrive in Osaka from Tokyo in less than around 3 hours. The pass will allow you to travel on the local JR train services for free, which will result in some transportation cost savings for you as travelling locally on the JR train is not that cheap. Northwest Airlines departs Singapore early in the morning, which means that you will arrive in Osaka late in the evening following arrival into Tokyo and taking the bullet train to Osaka. For your return flight, Northwest Airlines have a late flight out of Tokyo which allows you ample time to depart from Osaka into Tokyo Narita Airport.
